Melanotan I (MT-I), also known as Afamelanotide or NDP-α-MSH, is a synthetic 13-amino acid peptide analog of α-melanocyte-stimulating hormone (α-MSH). Two amino acid substitutions—norleucine at position 4 and D-phenylalanine at position 7—enhance its receptor affinity and improve resistance to enzymatic degradation compared with endogenous α-MSH.
MT-I functions primarily as an agonist of the melanocortin-1 receptor (MC1R), with activity at additional melanocortin receptor subtypes. Its receptor selectivity has made it an important research tool for investigating melanogenesis, melanocortin receptor pharmacology, pigment cell biology, and peptide-receptor interactions.
